Description: Ergocalciferol, also known as Vitamin D2 and calciferol, is a type of vitamin D found in food and used as a dietary supplement. As a supplement it is used to prevent and treat vitamin D deficiency. This includes vitamin D deficiency due to poor absorption by the intestines or liver disease. It may also be used for low blood calcium due to hypoparathyroidism. It is used by mouth or injection into a muscle.

More Info: Excessive doses can result in increased urine production, high blood pressure, kidney stones, kidney failure, weakness, and constipation. If high doses are taken for a long period of time, tissue calcification may occur. It is recommended that people on high doses have their blood calcium levels regularly checked. Normal doses are safe in pregnancy. It works by increasing the amount of calcium absorbed by the intestines and kidneys.[4] Food in which it is found include some mushrooms.

Biological target: Vitamin D2 (Ergocalciferol), drived from plant sources or dietary supplements, could be used as supplement of Vitamin D.
In vitro activity: Exploration of the acting mechanisms involved this event revealed that ergocalciferol induced DNA fragmentation and increased sub-G1 DNA contents in HL-60 cells, both of which are hallmarks of apoptosis. Analysis of the integrity of mitochondria demonstrated that ergocalciferol caused loss of mitochondrial membrane potential with release cytochrome c to cytosol, generation of reactive oxygen species (ROS), and depletion of glutathione (GSH), suggesting that ergocalciferol may induce apoptosis in HL-60 cells through a ROS-dependent pathway. In vivo activity: In the SNx group, ergocalciferol significantly increased overall contractile response to PE compared with vehicle (P < 0.0001) (Figure 3). The maximum achieved tension (g) was increased in the SNx group rats treated with ergocalciferol compared to SNx animals treated with vehicle whilst the potency remained unchanged (P = 0.03, Table 2).
