Description: Vesnarinone is a cardiotonic agent, mixed phosphodiesterase 3 inhibitor and ion-channel modifier that has modest, dose-dependent, positive inotropic activity, but minimal negative chronotropic activity. Vesnarinone improves ventricular performance most in patients with the worst degree of heart failure.

Biological target: Vesnarinone inhibits phosphodiesterase III (PDE3) activity.
In vitro activity: Downregulation of CXCR4 mRNA by treatment with vesnarinone was also detected in oral cancer cells, ACC-M, in a time dependent manner (Figure 2c). Moreover, the reduced expression of CXCR4 protein by the treatment with vesnarinone, as compared to the effects of 5-FU and CDDP both in ACC-M cells and in Hela cells (Additional File 1). SDF-1 was found to enhance the migration of B88 cells, which was in contrast significantly inhibited by treatment with vesnarinone (Figure 3a). In vivo activity: This study study demonstrated that vesnarinone inhibited the acute lung injuries, possibly by blocking the production of KC, and ameliorated the development of pulmonary fibrosis, since it is well known that the degree of the pulmonary fibrosis is dependent on the severity of initial lung injury. In 7 days after BLM treatment, elevated levels of cell numbers of all kinds of inflammatory cells, and protein and albumin contents in BALF were noted in CB group. Both low and high doses of vesnarinone intake significantly decreased all these levels. In addition, biochemical markers for lung inflammation in BALF including a neutrophilic chemokine, KC, sTNF-Rs, and hyaluronic acids, all which were increased in CB mice, were decreased in LB and HB mice.
