Biological target: | Esculin (Aesculin) is a glucoside found in horse chestnuts. |
In vitro activity: | RT-qPCR revealed that treating HepG2 cells with esculin (50, 100, and 200 µM) significantly restored the increase in TNF-α and IL-6 expression (Figure 5(D,E)). Similarly, this study confirmed that the expression of ac-NF-κB was markedly upregulated while that of Sirt1 was downregulated in the FFA-treated HepG2 cells. However, these changes were found to be ameliorated in esculin-treated cells (Figure 5(F–H)). Reference: Pharm Biol. 2021; 59(1): 922–932. https://www.ncbi.nlm.nih.gov/pmc/articles/PMC8274538/ |
In vivo activity: | Although esculin did not change metabolic parameters, renal biochemical function, neither TBARS in DM rats, esculin reduced P2X7 levels in these animals and restored mitochondrial function via glycolysis substrates and β-oxidation. Besides, at the histological analysis, this study observed that esculin reduced inflammatory infiltrates and collagen IV deposits as compared to diabetic group.
